BMD antituberculosis susceptibility
Rapidly growing mycobacteria (RAPMYCOI): 3–5 days incubation; final clarithromycin result at extended read up to 14 days. Slowly growing mycobacteria (SLOMYCOI): 7–21 days depending on species and growth rate.
Manual broth microdilution using dried Sensititre™ RAPMYCOI / SLOMYCOI 96-well MIC panels; visual reading of MIC against a growth-control well (CLSI M24 reference method).
Recommended by CLSI
Sensititre™ RAPMYCOI / SLOMYCOI broth microdilution MIC panels. Determination of minimum inhibitory concentrations (MIC) on confirmed NTM isolates by the CLSI reference method (CLSI M24), with interpretation per the current CLSI M24S supplement. Panel selection (species-dependent): RAPMYCOI for rapidly growing mycobacteria — M. abscessus complex, M. fortuitum group, M. chelonae; SLOMYCOI for slowly growing mycobacteria — M. avium complex, M. kansasii, M. marinum.RAPMYCOI agents: amikacin, amoxicillin-clavulanic acid, cefepime, cefoxitin, ceftriaxone, ciprofloxacin, clarithromycin, doxycycline, imipenem, linezolid, minocycline, moxifloxacin, tigecycline, tobramycin, trimethoprim sulfamethoxazole. SLOMYCOI agents: amikacin, ciprofloxacin, clarithromycin, doxycycline, ethambutol, ethionamide, isoniazid, linezolid, moxifloxacin, rifabutin, rifampin, streptomycin, trimethoprim-sulfamethoxazole. Reporting: S/I/R is issued only where CLSI M24S interpretive criteria exist for the identified species. For rapidly growing mycobacteria, clarithromycin is the macrolide-class surrogate and is read at extended incubation to detect inducible resistance; for M. avium complex, clarithromycin and amikacin are the agents with validated reportable breakpoints; for M. kansasii, clarithromycin and rifampin are primary. Agents without established criteria are reported as an MIC value with an explanatory comment, extra pulmonary samples may take up to 8-12 weeks of incubation as they are fastidious.

Broth microdilution (BMD) is the reference (gold-standard) method recommended by CLSI for testing antituberculosis susceptibility of Mycobacteria (such as MTB and MOTT).
Pure culture isolate of a rapidly or slowly growing NTM — a fresh subculture on Löwenstein–Jensen (LJ) slant or equivalent solid medium, with species identification already confirmed. Primary specimens are not accepted for this test.
Not applicable — a pure subculture with sufficient viable growth for a 0.5 McFarland suspension is required.
Screw-capped LJ slant or equivalent solid-medium tube, sealed and placed in secondary containment.
No special preparation is required for this test. You may eat, drink, and take your medications as normal, unless instructed otherwise by your healthcare provider.
Refrigerated temperature, upright, in leak-proof secondary packaging in accordance with UN 3373 Category B requirements.
